Albert Wesker made his first appearance as the primary antagonist in the original Resident Evil as the commander of the STARS and leader of the Alpha team. He appears through the course of the game as a character assisting the protagonist. As the player goes further into the game, evidence begins to appear in the mansion's underground laboratory implicating Wesker as a member of the Umbrella research staff. Wesker eventually reveals that he was ordered by his superiors at Umbrella to lure the STARS into the mansion, using them as test subjects against the mutated creatures to gather battle data.

Wesker returns as the main antagonist of Resident Evil 5 voiced by D.C. Douglas, who also voiced Wesker in the game "Resident Evil: The Umbrella Chronicles", and motion captured by Ken Lally.

IGN listed Wesker as one of their "Top 10 Memorable Villains" at number three, citing his role in the background of the series.
